Mechanism of Action (MOA):

  • NSAIDs acts primarily by inhibiting synthesis of prostaglandin; mediators involved in nociception and pathogenesis of inflammation and fever.
  • It blocks cyclooxygenase enzyme either reversibly or irreversibly.——————————————–> Inhibition of synthesis of prostaglandin, prostacyclin (PGI2), and thromboxane A2 (TXA2)

Pharmacological effects:

  1. Analgesia: It produce analgesic effect mainly by inhibiting production of prostaglandin and by decreasing sensitization of pain receptors to noxious stimuli.
  2. Antipyresis: NSAID reset the set point or thermostat; also increase heat loss through peripheral vasodilation by inhibiting synthesis of prostaglandin.
  3. Anti-inflammatory effect: NSAID mainly inhibit prostaglandin synthesis at site of injury and produce anti-inflammatory effects.
