 Endospore staining

Moeller stain: It is a differential stain which differentiates endospores forming and non-endospores forming bacteria. It is used to presence and location of endospores. Moeller stain uses carbol fuschin(red), acid-alcohol and methylene blue.

Steps:

  • Heat fix sample to the slide
  • Add carbol fuchsin. Then steam slide or add detergent.
  • Rinse with acid alcohol.
  • Add methylene blue as a counter stain.
