Culture media

Artificial culture media constitutes the food materials for the cultivation of microorganisms, which provides the in- vivo composition and conditions for the successful growth of bacteria, fungi and parasites in-vitro.

 

classification

  1. Based on consistency (state):
  • Liquid(no agar) : peptone water, nutrient broth
  • Semi-solid(1-1.5 % agar): nutrient agar stabs(TSI media)
  • Solid(2.5% agar): blood agar, serum agar

2 . Based on oxygen requirement:

  • Aerobic media: cultivation of aerobic organisms
  • Anaerobic media: cultivation of anaerobic organisms. Eg. Robertson’s cooked meat media, peptone infusion broth.

3 . Sabauround’s dextrose agar: use for the cultivation of fungi.

4 . Simple media:

  • It consists of only basic necessities and used for simply growth.
  • Peptone water, nutrient broth, nutrient agar
  • Peptone water(1% peptone+0.5%NaCl+100 ml water)
  • Nutrient broth(peptone water+1%meat extract)
  • Nutrient agar(nutrient broth+2% agar)
  • Used to grow non-fastidious microorganisms

5 . Complex media(special media):

  • Used to grow specific bacteria and constitute specific ingredients
  • Have added ingredients for special purpose for bringing out certain properties or for providing special nutrients for the growth of bacterium under study.
  • Enriched media, differential media, enrichment media, selective media, sugar media, transport media.
