Selection of Environment for TPS Production

• Long day condition is required to produce flower of potato
• For this, High hill and long day season is required.
• Season : rainy season
• Due to rain, hybridizing/crossing work is difficult, so parental line are planted in Plastic tunnel or glass house / green house condition which make the work easy
• However, in Terai and short day condition light from Sodium Tube Light is provided in crossing block from 5 : P M to 11 P M (in evening to night) for additional light
• For, 1 ropani land : 10 Sodium Tube Light is required.

Other practices

a) Collection of Male Flower

• While flowers are fully opened, before pollen dehisce, in morning before 11.00 a.m

b) Pollen extraction & storage

• Pollen are collected after one day of collection of flower. Flower are stored at room temperature for one day for easy collection of pollen.
• Pollen are collected by use of vibrator.
• Pollen are kept in air tight container at 4-10 oC temp for pollination.
• Pollen can be stored for 15 days if kept with the use of silica gel and desiccators.

c) Pollination Methods

• Stigma of female flower of female line is dipped in pollen kept in Petridis.
• Pollination by use of brush or forceps. Double pollination can be done within 24 hours.
• Pollination is done in morning because of high stigmatic receptivity.
