Detection of compounds on chromatogram
a) Physical method:
- Chromatogram observed first in daylight and the position of colored substance is marked.
- Chromatogram is then observed on ultraviolet light.
- Substance appear as fluorescent spots with characteristics hues.
b) Chemical method:
- Chromatogram is put on horizontal sheet of filter paper and sprayed with fine mist of reagent side by side.
- The substance appear as fluorescent substance.
c) Biological method:
- Chromatogram is pressed on a Petri dish with a corresponding culture of microorganism.
- Amylase detected by spraying the chromatogram with starch solution and iodine solution.
- Starch doesn’t react with iodine.
