Gel electrophoresis of DNA
- Used to identify, separate and purify DNA fragments.
- Location of DNA is identified by staining with ethidium bromide.
Steps:
- Agarose gels are cast by melting the agarose in the presence of desired buffer, until a clear, transparent solution is obtained.
- The melted solution is then poured into a mold and allowed to harden.
- Upon hardening, agarose form matrix.
- When electric field is applied across the gel, DNA which is -vely charged migrate towards the anode.
