Detection of compounds on chromatogram

a) Physical method:

  • Chromatogram observed first in daylight and the position of colored substance is marked.
  • Chromatogram is then observed on ultraviolet light.
  • Substance appear as fluorescent spots with characteristics hues.

 

b) Chemical method:

  • Chromatogram is put on horizontal sheet of filter paper and sprayed with fine mist of reagent side by side.
  • The substance appear as fluorescent substance.

 

c) Biological method:

  • Chromatogram is pressed on a Petri dish with a corresponding culture of microorganism.
  • Amylase detected by spraying the chromatogram with starch solution and iodine solution.
  • Starch doesn’t react with iodine.
